The varin family
CBDV stands in the same relationship to CBD that THCV does to THC. Three carbons on the side chain instead of five, everything else unchanged.
Like THCV, it comes from CBGVA rather than CBGA, so a plant that makes meaningful CBDV is running a different biosynthetic branch. High-CBDV chemotypes turn up in some landrace populations from India, Nepal and parts of Africa, and CBDV also appears at low levels in many CBD-dominant hemp cultivars.
What the trials found
CBDV attracted pharmaceutical attention because of anticonvulsant activity in animal seizure models. GW Pharmaceuticals, the company that developed Epidiolex, took it into human trials under the name GWP42006.
The adult focal seizure trial did not work. GW reported in 2018 that CBDV failed to beat placebo on seizure reduction, and the program in that indication stopped.
The autism work has been more mixed and is ongoing at academic centres rather than being driven by one company. Trials at Montefiore and elsewhere have looked at CBDV for irritability and repetitive behaviors in children with autism spectrum disorder, with modest and inconsistent findings.
Two things follow. CBDV has a stronger clinical evidence base than CBC, CBG or CBN. And that evidence base includes a clear failure, which is worth more than a marketing page will ever tell you. A negative trial is real information about a compound.
Mechanism
CBDV binds CB1 and CB2 weakly, which is the usual pattern for non-intoxicating cannabinoids. Its better-characterized activity is at TRPV1 and related TRP channels, where it acts as an agonist and then desensitizes the channel with continued exposure. Desensitization is the same phenomenon behind capsaicin creams: activate a sensory channel hard enough and it stops responding.
CBDV has also been reported to affect gene expression related to the endocannabinoid system in animal models. That work is early and has not been replicated in humans.
What this means for a shopper
The gap between the research and the shelf is wide. Trial doses ran into the hundreds of milligrams per day of pharmaceutical-grade isolate under medical supervision. A retail full-spectrum tincture might contain a few milligrams of CBDV per bottle, incidentally, alongside everything else.
Reading a clinical result and expecting it from a consumer product is the most common mistake in this category, and CBDV is where it happens most, because the research is genuinely more serious. The trials tell you about the molecule. The bottle is a separate question.
